QuantiFERON-TB Gold Blood Collection Guidelines
Collection Requirements
- The Centers for Disease Control and Prevention recommends that blood for the QuantiFERON-TB Gold test must be collected in heparinized vacutainer tubes to ensure proper interferon-gamma (IFN-γ) detection 1, 2, 3
- The test requires fresh heparinized whole blood for proper IFN-γ detection, as the heparin anticoagulant preserves white blood cell function, which is essential since the test measures IFN-γ released by sensitized T-cells after antigen stimulation 1, 3, 4
Collection Methods and Timing
- The FDA-approved direct collection into specialized QFT tubes (containing heparin plus specific antigens) is a recommended method, as it allows for immediate incubation with test antigens 6
- Blood must be incubated with test antigens within 12 hours of collection, as delayed processing compromises white blood cell viability and test accuracy 6
Important Practical Considerations
- The use of heparin anticoagulation is recommended, as it maintains cellular integrity without interfering with the immune response measurement, according to the Centers for Disease Control and Prevention 1, 2
- Other anticoagulants (EDTA, citrate) are not appropriate, as they can affect lymphocyte function 6
