Zaleplon Dosing and Efficacy Guidelines

Introduction to Zaleplon

  • The American Academy of Sleep Medicine recommends zaleplon primarily for sleep onset insomnia rather than sleep maintenance 1, 2
  • At the 10 mg dose, zaleplon demonstrates clinically significant reduction in sleep latency compared to placebo, with moderate strength of evidence 2

Dosing Considerations

  • The American Academy of Sleep Medicine suggests that for non-elderly adults, the recommended dose is 10 mg, with high strength of evidence 1
  • Elderly patients should receive 5 mg due to increased sensitivity to hypnotics, based on moderate strength of evidence 1

Safety and Efficacy

  • Treatment-emergent adverse events with zaleplon show no significant difference from placebo at recommended doses, with high strength of evidence 2, 4
  • Common adverse events include headache, asthenia, neurasthenia, pain, fatigue, and somnolence, based on moderate strength of evidence 4
  • No significant withdrawal symptoms or rebound insomnia have been observed following discontinuation of zaleplon at recommended doses, with moderate strength of evidence 4
