Midazolam Pharmacokinetics and Stability

Clinical Pharmacology

  • Midazolam's water solubility in acidic solution (pH 3) and its reconfiguration at physiologic pH (7.4) to increase lipid solubility contribute to its favorable pharmacokinetic profile 1, 2
  • The American Gastroenterological Association notes that when using midazolam clinically, be aware of its potential for respiratory depression, particularly when combined with opioids 1, 3

Special Populations

  • The drug's clearance is reduced in elderly patients, those who are obese, and patients with hepatic or renal impairment, which may necessitate dose adjustments regardless of storage conditions 3, 4
